چکیده

The inhibitor), effect of hop bract polyphenols (HBP) on cariogenic streptococci was investigated. It wu'as fbund that the high molecular weight polyphenol (estimated about 36,OeO-40,OOO) inhibited the cellular adherence of Streptoeoccus mutans MT8148 (serotype C) and Streptoeoeeus sobrinus ATCC 33478 (serotype g) at msuch smaller concentrations than the polyphenols extracted from oe)ong tea or green tea leayes, Fsurthermore, HBP also inhibited the actien ef glucosyltransferase, which was inyolyed in the waterinsoluble glucan s},nthesis, but did not suppress the growth and the acid production of the bacteria. These results suggest thait HBP would be a candidate to act against dental caries ¢ aused b)' Mutans Streptocoeci.

Characterization of an anti-glucosyltransferase serum specific for insoluble glucan synthesis by Streptococcus mutans.

An anti-glucosyltransferase serum, which synthesized 96% insoluble glucans, was prepared against a purified enzyme preparation from Streptococcus mutans strain HS6 (serotype a). This serum was examined for its effects on glucan synthesis by crude enzyme preparations from eight strains (four serotypes) of S. mutans and for the ability of these preparations to promote adherence of S. mutans to a ...

Effects of antibodies against cell surface protein antigen PAc-glucosyltransferase fusion proteins on glucan synthesis and cell adhesion of Streptococcus mutans.

Cell surface protein antigen (PAc) and glucosyltransferases (GTFs) produced by Streptococcus mutans are considered to be major colonization factors of the organism, and the inhibition of these two factors is predicted to provide protection against dental caries. In this study, we have constructed fusion protein PAcA-GB, a fusion of the saliva-binding alanine-rich region (PAcA) of PAc with the g...

Dextranases from oral bacteria: inhibition of water-insoluble glucan production and adherence to smooth surfaces by Streptococcus mutans.

The effect of dextranases (EC 3.2.1.11) from the oral isolates Actinomyces israelii and Bacteroides ochraceus on water-insoluble glucan production by the Streptococcus mutans dextransucrase (EC 2.4.1.5) and sucrose-dependent adherence to smooth glass surfaces by S. mutans was studied. Collection on membrane filters of water-insoluble polysaccharides synthesized from radioactive sucrose was used...

Recombination between gtfB and gtfC is required for survival of a dTDP-rhamnose synthesis-deficient mutant of Streptococcus mutans in the presence of sucrose.

The rml genes are involved in dTDP-rhamnose synthesis in Streptococcus mutans. A gene fusion between gtfB and gtfC, which both encode extracellular water-insoluble glucan-synthesizing enzymes, accompanied by inactivation of the rml genes was observed for cells grown in the presence of sucrose. The survival rates of rml mutants isolated in the absence of sucrose were drastically reduced in the p...

Purification and characterization of cell-associated glucosyltransferase synthesizing water-insoluble glucan from serotype c Streptococcus mutans.

Strains of Streptococcus mutans (serotypes c, e and f) were found to possess high levels of glucosyltransferase (GTase) activity, both cell-associated and in the culture medium, when grown in either sucrose-free or sucrose-containing broth media. The cell-associated GTase of S. mutans MT8148 (serotype c) was effectively extracted by treatment with 8 M-urea at 25 degrees C for 1 h. Approximately...
